Architecture and relevance of several strongly adhered biofilms over a polyester imide (PEI) surface BJM
Soncini-Júnior,Giancarlo; Franchetti,Sandra Mara Martins; Marconato,José Carlos.
A microscopical study was undertaken to analyze the eventual fungi adhesion over a polyester imide surface in enameled copper wires. Scanning electron microscopy allowed to observe in these adhered biofilms, a high amount of pigments, hyphae and its enzymatic arsenal probably acting in the surface of the polymer. Due to the highly aromatic nature of this polyester and the residual phenol derivates still present in the reticulated polymer, an antifungal activity could be expected, but no significant changes were noted in fungal growing and in the adhesion process. Additionally, the wires showed completely failure in all insulation properties. The studies aimed to understand and evaluate the great fungi potential that could be explored in biodeterioration...

Bioprotective Effect of Lactococcus piscium CNCM I-4031 Against Listeria monocytogenes Growth and Virulence ArchiMer
Saraoui, Taous; Leroi, Francoise; Chevalier, Frederique; Cappelier, Jean-michel; Passerini, Delphine; Pilet, Marie-france.
Listeria monocytogenes is a Gram-positive pathogen occurring in many refrigerated ready-to-eat foods. It is responsible for foodborne listeriosis, a rare but severe disease with a high mortality rate (20–30%). Lactococcus piscium CNCM I-4031 has the capacity to prevent the growth of L. monocytogenes in contaminated peeled and cooked shrimp and in a chemically defined medium using a cell-to-cell contact-dependent mechanism. To characterize this inhibition further, the effect of L. piscium was tested on a collection of 42 L. monocytogenes strains. All strains were inhibited but had different sensitivities. The effect of the initial concentration of the protective and the target bacteria revealed that the inhibition always occurred when L. piscium had reached...

Chemical-Mineralogical Characterization of Magnetic Materials from Magnetic Soils of the Southern Espinhaço Mountain Chain and of the Upper Jequitinhonha Valley, State of Minas Gerais, Brazil Rev. Bras. Ciênc. Solo
Silva,Alexandre Christófaro; Souza,Solange de; Fabris,José Domingos; Barbosa,Maurício Soares; Barral,Uidemar Morais; Costa,Roberto Vial.
ABSTRACT In the Southern Espinhaço Mountain Chain and in the Upper Jequitinhonha Valley, magnetic soils, in different pedogenetic stages, are found to be forming over intrusions of basic lithology. The essential chemical and mineralogical properties of samples from magnetic soil profiles from those two physiographic environments in the state of Minas Gerais, Brazil, are reported. Three of the pedons (Rhodic Kandiustox – RKox, Rhodic Haplustox – RHox, and Typic Argiustoll - TAoll) were identified as being indeed developed over basic rocks; the fourth pedon (Typic Haplustox - THox) is currently forming on an acidic rock. Particle size and routine chemical analyses were performed on samples from all horizons of the four selected soil profiles. For a deeper...

Citrus sinensis leaf petiole and blade colonization by Xylella fastidiosa: details of xylem vessel occlusion Scientia Agricola
Alves,Eduardo; Leite,Breno; Pascholati,Sérgio Florentino; Ishida,Maria Lúcia; Andersen,Peter Craig.
Citrus variegated chlorosis (CVC), caused by Xylella fastidiosa, is an important disease of citrus in Brazil. X. fastidiosa is restricted to xylem vessels of plants and knowledge regarding xylem colonization is still limited. Our goal was to verify how this bacterium colonizes and spreads within xylem vessels of sweet orange Citrus sinensis cv. Pêra. Petioles and pieces of leaf blades from naturally infected plant exhibiting characteristic symptoms were prepared for light microscopy (LM), scanning electron microscopy (SEM), transmission electron microscopy (TEM) and immunogold labeling (IGL). Petioles from healthy plants were used as control. IGL results, using an antibody against wall hemicelluloses, revealed that the pit membrane of vessels was altered....

Comparative leaf anatomy of some species of Abies and Picea (Pinaceae) Acta Botanica
Ghimire,Balkrishna; Lee,Chunghee; Yang,Jongcheol; Heo,Kweon.
ABSTRACTA number of conifer species are still lacking anatomical data, which is significant because morphological and anatomical data are essential for systematic study. Leaf anatomy was studied in selected species of Abiesand Piceausing light and scanning electron microscopy. Both genera were found to have typical coniferous and highly xerophytic leaves with sunken stomata and an epidermis covered by a thick cuticle. In the genus Abies, species can be differentiated by the nature of the lignified hypodermis and the number and position of resin ducts. Abies firma and A. holophylla have a continuous hypodermis whereas in A. koreana and A. nephrolepis the hypodermis is discontinuous and represented by isolated cells or groups of four or five cells. On the...

Comparative Wood Anatomy of the Primuloid Clade (Ericales s.l.) Naturalis
Lens, F.; Jansen, S.; Caris, P.; Serlet, L.; Smets, E..
The wood structure of 78 species from 27 genera representing the woody primuloids (Maesaceae, Myrsinaceae, and Theophrastaceae) was investigated using light microscopy (LM) and scanning electron microscopy (SEM). Results indicated that the ray structure, the nature of mineral inclusions, and the occurrence of breakdown areas in rays can be used to separate the three primuloid families from each other. Within Ericales, the presence of exclusively multiseriate rays is synapomorphic for Myrsinaceae and Theophrastaceae, and the occurrence of breakdown areas in rays is synapomorphic for Myrsinaceae. Within Myrsinaceae, the wood structure of the mangrove genus Aegiceras differs because it has short vessel elements that are storied, non-septate fibers, a...

Comparison of the physical and geotechnical properties of gas-hydrate-bearing sediments from offshore India and other gas-hydrate-reservoir systems ArchiMer
Winters, W. J.; Wilcox-cline, R. W.; Long, P.; Dewri, S. K.; Kumar, P.; Stern, L.; Kerr, L..
The sediment characteristics of hydrate-bearing reservoirs profoundly affect the formation, distribution, and morphology of gas hydrate. The presence and type of gas, porewater chemistry, fluid migration, and subbottom temperature may govern the hydrate formation process, but it is the host sediment that commonly dictates final hydrate habit, and whether hydrate may be economically developed. In this paper, the physical properties of hydrate-bearing regions offshore eastern India (Krishna-Godavari and Mahanadi Basins) and the Andaman Islands, determined from Expedition NGHP-01 cores, are compared to each other, well logs, and published results of other hydrate reservoirs. Properties from the hydrate-free Kerala-Konkan basin off the west coast of India are...

Cytokinin preconditioning enhances multiple shoot regeneration in Pongamia pinnata (L.) Pierre - a potential, non-edible tree seed oil source for biodiesel Electron. J. Biotechnol.
Belide,Srinivas; Sajjalaguddam,Ramgopal Rao; Paladugu,Anitha.
An efficient, highly reproducible protocol for multiple shoot induction and plant regeneration of Pongamia pinnata has been successfully developed using cotyledonary node explants. This study also demonstrates that preconditioning of explant stimulates production of multiple shoots from cotyledonary nodes of P. pinnata. The highest direct shoot regeneration (90%) with an average of 18.4 3.1 shoots/explant were obtained when cotyledonary node explants were excised from seedlings germinated on Murashige and Skoog (MS) media supplemented with benzyladenine (BA) 1 mg l-1, and subsequently cultured on MS media with 1 mgl-1 thidiazuron (TDZ). Scanning electron microscope observations of cotyledonary node (CN) explants excised from pre-conditioned and normal...

Ecophysiological, anatomical and ultrastructural characteristics of Vitex polygama Cham. (Verbenaceae) submitted to different concentrations of fluoride Biological Sciences
Rossi, Zélia Terezinha Teixeira; Castro, Evaristo Mauro de; Pereira, Giuslan Carvalho; Barbosa, João Paulo Rodrigues Alves Delfino; Carvalho, Graziele Wolff de Almeida; Rossi, Marco Aurélio de Lima Silva; Pereira, Fabrício José.
Fluorides released during the manufacture of aluminum, fertilizers, glass and ceramics are considered to be highly toxic to plants, causing damages in relatively small concentrations. This study aimed to evaluate fluoride (F) accumulation and its effects on Vitex polygama Cham. (Verbenaceae) exposed to simulated rain containing potassium fluoride in concentrations of 5, 10, 15 and 20 mg L-1. Vitex polygama presented no visual symptoms of foliar injury and accumulated a relatively small amount of F-, significant only for the treatments with higher concentrations of the pollutant.  F- promoted an increase in non-photochemical quenching (qN) and in the coefficient due to non-photochemical extinction (NPQ), which indicates higher dissipation of radiant energy...

Effect of fungicide on Fusarium verticillioidesmycelial morphology and fumonisin B1 production BJM
Miguel,Tatiana de Á.; Bordini,Jaqueline G.; Saito,Gervásio H.; Andrade,Célia G.T. de J.; Ono,Mario A.; Hirooka,Elisa Y.; Vizoni,Édio; Ono,Elisabete Y.S..
The effect of fludioxonil + metalaxyl-M on the mycelial morphology, sporulation and fumonisin B1 production by Fusarium verticillioides 103 F was evaluated. Scanning electron microscopy analysis showed that the fungicide caused inhibition of hyphal growth and defects on hyphae morphology such as cell wall disruption, withered hyphae, and excessive septation. In addition, extracellular material around the hyphae was rarely observed in the presence of fludioxonil + metalaxyl-M. While promoting the reduction of mycelial growth, the fungicide increased sporulation of F. verticillioides compared to the control, and the highest production occurred on the 14th day in the treatments and on the 10th day in the control cultures. Fumonisin B1production in the culture...

Effect of morphological heterogeneity of somatic embryos of Melia azedarach on conversion into plants Biocell
Vila,Silvia; Gonzalez,Ana; Rey,Hebe; Mroginski,Luis.
Embryogenic cultures were initiated from immature Melia azedarach (Meliaceae) zigotic embryos. Explants were induced on Murashige and Skoog (1962) medium with 4.54 μM thidiazuron or 0.45 μM dichlorophenoxyacetic acid. After 6 weeks of culture on induction medium, somatic embryos were categorized in four morphological classes based on the presence of single or fused embryos and if they remained united or not to the original explant; that were evaluated histologically. The somatic embryos of every category were transferred, in groups or individually, on a 1/4 MS medium. Bipolar embryos, the more typically normal ones, had well defined shoot and root apical meristems and produced single plants; subcultured individually their conversion was...

Effect of ultrasonication of Switchgrass on fermentable sugar production and biomass physical structure CIGR Journal
Sharma, Rajat; Sharma-Shivappa, Ratna R.; Boyette, Michael D.; Stikeleather, Larry F..
Pretreatment of lignocellulosic biomass for enhancing sugar yields has been studied extensively over the years.  Conventional methods employed to preprocess biomass to make it conducive for enzymatic hydrolysis have been hampered by issues like poor energy efficiency and production of undesirable by-products.  Ultrasonication, which involves the treatment of biomass through ultrasonic sound waves in a liquid medium without additional chemicals, is believed to have potential for biomass pretreatment.  In this study the effects of ultrasonication on switchgrass, a potential feedstock for bioethanol production due to its high cellulosic content, were investigated.  Results of compositional analysis and scanning electron microscopy conducted to visualize...

Etude du biofilm microalgal des vasières intertidales : dynamique spatio-temporelle à micro-échelle et performances photosynthétiques ArchiMer
Herlory, Olivier.
The microalgal biofilm, which is present on sediment surface, is the primary production unit on bare intertidal mudlfats. In order to improve our understanding of this primary production system, a study was undertaken at microscale to analyse the structural and functional properties of this biofilm. In the first part of this work, a detailed descriptive analysis lead to the characterisation of the spatiotemporal dynamics of the physical structure of the biofilm. It turns out that the biofilm exhibits its own dynamics at the sediment surface (as a discrete compartment in the top 200 µm), but still has a functional relationship with the total available biomass (PCB) distributed within the top 1 cm (linear relationship between the maximum size of the biofilm...

External development of the entomopathogenic fungi Beauveria bassiana and Metarhizium anisopliae in the subterranean termite Heterotermes tenuis Scientia Agricola
Moino Jr.,Alcides; Alves,Sérgio Batista; Lopes,Rogério Biaggioni; Neves,Pedro Manuel Oliveira Janeiro; Pereira,Roberto Manoel; Vieira,Solange Aparecida.
The subterranean termite Heterotermes tenuis is one of the main pests of sugarcane and eucalyptus in Brazil, and the use of entomopathogenic fungi, alone or associated to chemicals, is an efficient and environmentally favorable method for its control. Studies related to the fungal development on these insects are important due to the effect of insect behavior on entomopathogens. The objective of this work was to describe the external development of Beauveria bassiana and Metarhizium anisopliae on H. tenuis using Scanning Electron Microscopy (SEM), determining the duration of the different phases of fungal infection. Two fixation techniques for preparing SEM samples were also evaluated. Worker specimens of H. tenuis were inoculated with a 1 x 10(9) conidia...
